Nanotechnology meets radiobiology: Fullerenols and Metallofullerenols as nano-shields in radiotherapy

Despite advances in radioprotective measures, clinical use remains limited due to toxicity and low efficacy. In radiotherapy, conventional agents may also reduce treatment effectiveness. Fullerenols (FulOHs) and metallofullerenols (Me@FulOHs) offer a promising alternative with superior antioxidant properties and reduced pro-oxidative risks under optimized conditions. Me@FulOHs also serve as theranostic tools, enhancing PET/MRI imaging for early cancer detection. Their dual role in protection and diagnosis supports safer, more precise radiotherapy. This review explores their potential, alongside ionizing radiation effects, classifications, and countermeasures.
